Quantitative analysis of purine and pyrimidine nucleosides and bases in urine

Abstract

Background: the concentration of purine and pyrimidine metabolites in urine is useful marker of defects in their biosynthetic pathways or in the urea-cycle. Matherials and Methods: urine were collected from 58 healthy subjects aged from <2years to 42 years. The separation was performed with HPLC method. The same urine sample were examined using 1H-NMR. Results: HPLC and 1H-NMR method allowed the identification of 13 purine and pyrimidine compounds (orotic acid, creatinine, uracil, uric acid, hypoxantine, xanthine, uridine, thymine, inosine, guanosine, deoxyinosine, deoxyguanosine, deoxyadenosine). For each compound we have established reference ranges of concentration. The results point out that some compounds show a different urinary excretion related to age.
